Anthracnose Resistance

Markers for Anthracnose Resistanc

The Australian narrow-leafed lupin (Lupinus angustifolius) cultivars Tanjil and Wonga contain a single dominant gene Lanr1 conferring anthracnose resistance. Initially marker “AntjM1" was developed and implemented in the lupin breeding program. However marker AntjM1 can only be used for marker assisted selection (MAS) in a limited number of crosses due to genetic recombinations.  Recently, marker “AntjM2” was developed, which has been used in a wide range of crosses.

DNA sequence of marker “AntjM1”:

5’-CCCA(TTG)n*GTTTGTGC  ATTAGCATTT  GGTATTGGAA  ATGACTTGAC  AAATTCTGAA  GTTTCTTTGA  GATTGAGCTT  GCAAAGTGGA  TCAGGGAAAT  CATCTCCAAA  AGGGTTGTTT  TTGTTTTTCC  CATACAACCC  CATGTTGCTC  TCAAAGGTTG  AATCCTACAT  AATAATAATA  ATAAAACACA  AAAGAAAAAT  GAGAAAAGAA  AAGAAAAACA  TCTTGTAATA ATGCATAACA  AATGATATGC  TTCATATGTG  AGGATGGTTC  ATTGATTCTT  AGATTCTTAC  CATATGTGTT  GTTAA

*The sequence contains a microsatellite sequence (TTG)n.  The resistance allele has (TTG)6.  We found two susceptible alleles, one allele has (TTG)7, and the other has (TTG)4.  In another word, the susceptible band can be 3 bp longer, or 6 bp shorter than the resistant band.  For more information, please contact Dr. H. Yang ([email protected]).

DNA sequence of marker “AntjM2”:

Resistant allele: 5’- GTACCTGATG  ACAATTAGTC  ACACTTAGTA    ATGAGTTTGA  CATAAGTTGC  ATTAGGGTCA  TAACCACATT  ATTACTATAG  ACTAAAACCA  ATTTTTTTAA  CAAAAAAACA  TCAAAACCAA  AGGAATTTTA  TAAAGACTAA  ATTTCCTGGA  ACAAAAAACA  GATTTAAACC  TAGCAAAATT  AATAATTTAC  AGTTACACCA  AGACAAAAGA  AAGTAGAAAA  TATCATTGCA  GAAAAGGAGC  CTCCCGTAAA  ACATGCACAC  GCATCAAACT  AAGGAACAAG  GATCACTTGT  CCCAAAGTCT  GTCCAAATTA  TCTGTCCTCA  CCCATTAAAA  ATTGACACAT  GAGTAAATAG  AG[T]TTTTTTA  AAAACAAAAT  GATATATTTT  TATA[TTAC]TT  GTTCACAGTT  GATTGGACTG  AGATAGGATT  TAGAGATGAG  ATGTGGGATA  GAGGATCTCC  TTCCCAAACT  AAGAGGATGG  GGACAACTAT  GGATCCTTCC  ATTGGTTTTA  TCTAGGAGTG  AACATGACTC

Susceptible allele: Three susceptible alleles were found. One allele is 1 bp shorter (one base of “T” deletion), one is 4 bp shorter (“TTAC” deletion), and the other is 5 bp shorter (deletion at both sites) than the resistance allele.
